The patent situation
Tenofovir Alafolamine compound patent CN200410097845, expires in July 2021. Patent reported the preparation method of TAF, through adenine and R - the propylene carbonate reaction intermediates 1, again with p-toluene sulfonyl oxygen ethyl methyl con alkylation anti Chemicalbook should generate the intermediate 2, then add three methyl bromide silane hydrolysis yield of 3, again with phenol condensation, control of reaction conditions to form single phenyl phosphate intermediates 4, the use of sulfoxide chloride converting hydroxyl chlorine atom, again with L - alanine isopropyl ester N - alkylation reaction, finally resolution to get the target product.
An improved synthetic method is reported on CN201280048965, and the main difference is that under the same synthetic conditions as those mentioned above, intermediate 5 is generated from intermediate 1 in Chemicalbook basis with parathoxy, and intermediate 4 is obtained from LiOH monohydrolyzed phenyl phosphate.
