Pharmacology and toxicology
Lamivudine is a nucleoside antiviral drug, which has a strong inhibitory effect on hepatitis B virus (HBV) in vitro and in experimental infected animals.
Lamivudine metabolizes lamivudine triphosphate in HBV-infected cells and normal cells, which is the active form of Lamivudine and is both an inhibitor of HBV polymerase and a substrate for this polymerase. Lamivudine triphosphate was added to the viral DNA strand to block the synthesis of viral DNA. Lamivudine triphosphate does not interfere with normal cell deoxyribonucleoside metabolism, and it has little effect on mammalian DNA polymerase and, and has little effect on mammalian cell DNA content.
Lamivudine had no obvious toxicity to mitochondrial structure, DNA content and function. Serum HBV DNA test results in most hepatitis B patients showed that lamivudine rapidly inhibited HBV replication, and its inhibitory effect continued throughout the treatment process. At the same time, the serum transaminase can be reduced to normal level. Long-term application can significantly improve the inflammatory changes of liver necrosis and reduce or prevent the progression of liver fibrosis.
