1. Dissolve diethyl phosphite, paraformaldehyde and triethylamine in toluene, react under the protection of nitrogen, and reflux. After the reaction is complete, it is cooled, and p-toluenesulfonyl chloride and triethylamine are slowly added to react to obtain diethyl p-toluenesulfonyloxymethylphosphonate. Under the protection of nitrogen, (Y)-glycidol, 5% palladium-carbon, ethanol and sodium hydroxide aqueous solution are mixed and hydrogenated to obtain (R)-1,2-propanediol. Add the ethanol solution of diethyl carbonate and sodium ethoxide to react to obtain (R)-1,2-propylene glycol carbonate. Dissolve it with adenine, diethyl p-toluenesulfonyloxymethylphosphonate, and sodium hydroxide in dimethylformamide, and react to obtain (R)-9-[2-(diethylphosphonomethoxy)) Propyl] Adenine. Dissolve it in acetonitrile, add bromotrimethylsilane to obtain crude tenofovir, and recrystallize to obtain pure product. Mix tenofovir, 1-methyl-2-pyrrolidone, and triethylamine into chloromethyl propyl carbonate for reaction, and then acidify with fumaric acid to obtain tenofovir fumarate double pivaloyloxy group Methyl ester.
